چکیده

Purpose Merkel cell carcinoma (MCC) is a highly aggressive skin cancer with strong evidence for viral carcinogenesis. The association of MCC with the Merkel cell polyomavirus (MCPyV) may explain the explicit immunogenicity of MCC. Indeed, MCPyV-encoded proteins are likely targets for cytotoxic immune responses to MCC as they are both foreign to the host and necessary to maintain the oncogenic phenotype. However, to date only a single MCPyVderived CD8 T-cell epitope have been described, thus impeding specific monitoring of T-cell responses to MCC.

Discovery of CD8 T cell epitopes in Merkel cell polyomavirus through combinatorial encoding with MHC multimers

Merkel cell carcinoma (MCC) is a highly aggressive skin cancer induced through oncogenic transformation by the Merkel cell polyomavirus (MCPyV). Intratumoral CD8 T cell infiltration and the immunogenic capacity of the host have been strongly correlated to survival in patients with MCC. The MCPyV oncoproteins represent highly attractive targets for immunotherapy of MCC, being both foreign to the...

Preclinical evaluation of NF-κB-triggered dendritic cells expressing the viral oncogenic driver of Merkel cell carcinoma for therapeutic vaccination

BACKGROUND Merkel cell carcinoma (MCC) is a rare but very aggressive skin tumor that develops after integration of a truncated form of the large T-antigen (truncLT) of the Merkel cell polyomavirus (MCV) into the host's genome. Therapeutic vaccination with dendritic cells (DCs) loaded with tumor antigens is an active form of immunotherapy, which intends to direct the immune system towards tumors...

Merkel Cell Polyomavirus: A Causal Factor in Merkel Cell Carcinoma

The RNA viruses hepatitis C virus and human T-cell lymphotropic virus type I (or human T-cell leukaemia virus type 1), and the DNA viruses hepatitis B virus, high-risk human papillomaviruses, human herpes virus-8 (Kaposi’s sarcoma-associated herpes virus) and Epstein Barr virus are acknowledged as carcinogenic to humans. Human polyomaviruses have oncogenic potentials in cell cultures and animal...
